Minor Histocompatibility Antigen HA-1 Frequency and Acute GVHD in Some Hematopoietic Stem Sell Transplanted Iranian Patients

چکیده مقاله:
Background: It is suggested that HA-1 mismatching among hematopoietic stem cell transplanted recipients-donors is associated with acute graft-versus-host disease (aGVHD). So the aim of this study was to evaluate HA-1 frequency and examine the correlation between HA-1 disparity and GVHD patients who received transplantation from a HLA-identical sibling. Samples and Methods: the DNA was extracted from 30 pairs of HLA-A2-positive Iranian recipients-donors with GVHD I-IV and 25 pairs without GVHD. All the patients received HSCT from HLA-identical siblings. HA-1 was detected by SSP-PCR method. The HA-1 typing was performed using SSP method (SSP Minor Histocompatibility Antigen primer sets). Results: The frequency of HA-1R and HA-1H alleles in patients were 0.55 and 0.45 respectively, and showed no significant difference with these alleles frequency in donors 0.53 and 0.47 respectively (p>0.05). The HA-1 disparity was detected in 8 out of 55 donor/recipient pairs (14.5%). The aGVHD (grades I-IV) occurred in 6 (75%) out of 8 patients but in spite of higher incidence of it in the group of patients with HA-1 incompatibility, it was not statistically significant. Conclusion: In spite of higher frequency of HA-1 disparity in GVHD+ group our data did not reflect a significant association between the HA-1 disparity and risk of acute GVHD.

متن کاملThe Hematopoietic System–specific Minor Histocompatibility Antigen HA-1 Shows Aberrant Expression in Epithelial Cancer Cells
Allogeneic stem cell transplantation (SCT) can induce curative graft-versus-tumor reactions in patients with hematological malignancies and solid tumors. The graft-versus-tumor reaction after human histocompatibility leukocyte antigen (HLA)-identical SCT is mediated by alloimmune donor T cells specific for polymorphic minor histocompatibility antigens (mHags). Among these, the mHag HA-1 was fou...
متن کاملCord blood comprises antigen-experienced T cells specific for maternal minor histocompatibility antigen HA-1.
Umbilical cord blood transplantation is applied as treatment for mainly pediatric patients with hematologic malignancies. The clinical results show a relatively low incidence of graft-versus-host disease and leukemia relapse. Since maternal cells traffic into the fetus during pregnancy, we questioned whether cord blood has the potential to generate cytotoxic T cells specific for the hematopoiet...
متن کاملDoes minor histocompatibility antigen HA‐1 disparity affect the occurrence of graft‐versus‐host disease in tunisian recipients of hematopoietic stem cells?
INTRODUCTION Minor histocompatibility antigen HA-1 (MiHAg-HA-1) disparity between a patient and his or her human leukocyte antigen (HLA) genoidentical donor has been widely associated with an increased risk of graft-versus-host disease following allogeneic hematopoietic stem cell transplantation. OBJECTIVE To examine the effect of HA-1 disparity on the incidence of both acute and chronic graf...
متن کاملCD8(+) minor histocompatibility antigen-specific cytotoxic T lymphocyte clones eliminate human acute myeloid leukemia stem cells.
Effective immunotherapy for human leukemia based on infusions of T lymphocytes requires the identification of effector T cells that target the leukemic stem cell. The transplantation of human acute myeloid leukemia into nonobese diabetic/severe combined immune deficient (SCID) mice has identified a rare leukemic progenitor termed the SCID leukemia-initiating cell, which is present in low freque...

Recipient disparity for the HA-1 minor histocompatibility antigen has been associated with acute graft-versus-host disease (GVHD) after marrow transplantation from an HLA-identical sibling.1 We have also shown a trend toward higher risk of grades II to IV acute GVHD in recipients mismatched for HA-1 among a selected HLA-A2–positive population (P 5 .08), but the odds ratio (2.1) was lower than t...
